And lest you think this product is just for women, know that influencers like Andrew Huberman, Brian Johnson, and Gary Brecka also endorse its use.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Nicotinamide adenine dinucleotide (NAD+) has been one of the hottest anti-aging supplements in recent years. It&#8217;s sold by retail giants like Walgreens and Walmart, as well as trendy supplement companies.<\/p>\n<p class=\"css-ac37hb evys1bk0\">This molecule is important for cellular metabolism and other fundamental biological processes, and scientists and supplement companies alike claim that its levels decline with age. Despite the fact that evidence of benefit in humans is sparse, the idea that taking NAD+ supplements may aid in healthy aging has spread.<\/p>\n<p class=\"css-ac37hb evys1bk0\">And new research suggests that some of the thinking that fueled the boom in these supplement markets may be wrong. A study published this month in Nature Metabolism showed that blood levels of NAD+ do not actually decline with age. Another recent study, published as a preprint paper in February and not yet peer-reviewed, reached the same conclusion.<\/p>\n<\/div>\n<aside class=\"css-ew4tgv\" aria-label=\"companion column\"\/><\/div>\n<div data-testid=\"companionColumn-1\">\n<div class=\"css-53u6y8\">\n<p class=\"css-ac37hb evys1bk0\">&#8220;It&#8217;s become kind of a myth that NAD+ levels universally decline with age and that this contributes to age-related functional decline,&#8221; said Matt Kaeberlein, an associate professor at the University of Washington who studies NAD+ but was not involved in the new study. &#8220;This should make people question how real it is.&#8221;<\/p>\n<p class=\"css-ac37hb evys1bk0\">In the latest study, researchers analyzed blood samples from seven datasets totaling more than 300 people. Some of the data compares young people and older people. Other data have been obtained from frail individuals and elite athletes. The study showed that supplements can increase NAD+ levels, but found no evidence that they decrease with age.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Riekert Houtkooper, professor of translational metabolism at the University Medical Center Amsterdam, who led the study, said his team originally conducted the study because they wanted to develop a biomarker test to help people decide whether they should take NAD+ supplements. They were &#8220;very disappointed in the outcome,&#8221; he said.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Not everyone is disappointed with the findings. Joseph Bauer, a professor of physiology at the University of Pennsylvania who was not involved in the study, said that while knowing what happens with blood levels of NAD+ is valuable, &#8220;I don&#8217;t think it changes things very much&#8221; when it comes to basic theories about NAD+ and aging.<\/p>\n<p class=\"css-ac37hb evys1bk0\">That&#8217;s because several other human studies have found that NAD+ levels actually decrease in certain tissues, such as muscle and the brain, Dr. Baur said. (Dr. Haut-Cooper published one such study in 2022.) However, Dr. Baur acknowledged that the study is still &#8220;preliminary.&#8221;<\/p>\n<\/div>\n<aside class=\"css-ew4tgv\" aria-label=\"companion column\"\/><\/div>\n<div data-testid=\"companionColumn-2\">\n<div class=\"css-53u6y8\">\n<p class=\"css-ac37hb evys1bk0\">The bigger question, Dr. Bauer said, is whether age-related tissue decline is sufficient to cause functional impairment. Experts know that extremely low NAD+ levels can cause serious health problems. However, normal aging is associated with a much smaller decline, by some estimates around 30%. And a study in mice published last year found that reducing intramuscular NAD+ levels by as much as 85% did not seem to affect muscle function.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Andrew Hsiao, senior vice president of global regulatory and scientific affairs at Niagen Biosciences, which sells NAD+ supplements, said the new research &#8220;changes very little&#8221; for him. Like Dr. Bauer, he believes that studies showing a decrease in NAD+ in tissues are more important.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The theory that NAD+ declines with age emerged about 20 years ago, and studies in yeast, worms and mice have shown that increasing the molecular weight of NAD+ can extend lifespan, Dr. Koeberlein said. But testing NAD+ blood levels is technically very difficult, so until recently scientists were unable to study whether the decline occurs in humans, he added.<\/p>\n<p class=\"css-ac37hb evys1bk0\">But the theory took off and &#8220;outperformed the actual data,&#8221; Dr. Koeberlein said. &#8220;Prominent people have written reviews, spoken at conferences, and recently appeared on podcasts, describing what I call early-stage models as if they were proven fact.&#8221;<\/p>\n<p class=\"css-ac37hb evys1bk0\">&#8220;And as soon as it entered the supplement space, the need for data went out the window and everything became a marketing exercise,&#8221; he continued.<\/p>\n<\/div>\n<aside class=\"css-ew4tgv\" aria-label=\"companion column\"\/><\/div>\n<div data-testid=\"companionColumn-3\">\n<div class=\"css-53u6y8\">\n<p class=\"css-ac37hb evys1bk0\">Dr. Hsiao acknowledged that &#8220;the messages to consumers and the messages spread by influencers out there are probably oversimplified.&#8221; He added: &#8220;If we say that NAD+ levels are decreasing with age, we probably need to do some education to make it clear that what the evidence shows is happening in the tissues, not the blood.&#8221;<\/p>\n<p class=\"css-ac37hb evys1bk0\">Dr. Hout-Cooper said that while he has not given up on the therapeutic potential of NAD+, he sees no reason for normal, healthy people to take NAD+ as a supplement. Rather than focusing on aging in general, he studies whether aging is beneficial for people with rare genetic diseases that affect cellular metabolism.<\/p>\n<p class=\"css-ac37hb evys1bk0\">&#8220;There is still a lot of potential in the field of NAD+,&#8221; said Dr. Hout-Cooper.
